چکیده
Bromopyruvate inactivates the pyruvate dehydrogenase complex of Escherichia coli in a thiamine pyrophosphate (TPP)-dependent process. The catalytic activities of the individual enzyme components within the complex are not destroyed by bromopyruvate under similar conditions, but the activities of the pyruvate dehydrogenase and dihydrolipoyl dehydrogenase components are reduced in thiamine pyrophosphate-independent processes. Radioactivity from [2J4C]bromopyruvate is irreversibly bound in the presence or absence of thiamine pyrophosphate, but the amount of radioactivity bound in the presence of thiamine pyrophosphate is 3.9 times the amount bound in its absence. The data are consistent with the interpretation that TPP-dependent inactivation involves the action of bromopyruvate as an irreversible inhibitor which is directed into the complex by specific, but not necessarily irreversible, interaction with the pyruvate dehydrogenase component and ultimately blocks some site within the complex which is essential for catalytic activity in the over-all reaction but not in partial reactions catalyzed by the component enzymes.

Manganese (?) complex with 2, 2? bipyridine, 1, 1? dioxide (bpdo) ligand was immobilized within nanoreactors of MCM-41. The immobilized complex was characterized by powder x-ray diffraction (XRD), nitrogen adsorption desorption, and FTIR. The pore volume, surface area and pore diameter of MCM-41 decreased after immobilization of Mn complex. Three bands observed at 1250, 1220 cm?1 and 757 cm?1 a...
متن کاملCharacterization of the specific pyruvate transport system in Escherichia coli K-12.
A mutant of Escherichia coli K-12 lacking pyruvate dehydrogenase and phosphoenolpyruvate synthase was used to study the transport of pyruvate by whole cells. Uptake of pyruvate was maximal in mid-log phase cells, with a Michaelis constant for transport of 20 microM. Pretreatment of the cells with respiratory chain poisons or uncouplers, except for arsenate, inhibited transport up to 95%. Lactat...
متن کاملEnzymological and physiological consequences of restructuring the lipoyl domain content of the pyruvate dehydrogenase complex of Escherichia coli.
The core-forming lipoate acetyltransferase (E2p) subunits of the pyruvate dehydrogenase (PDH) complex of Escherichia coli contain three tandemly repeated lipoyl domains although one lipoyl domain is apparently sufficient for full catalytic activity in vitro. Plasmids containing IPTG-inducible aceEF-IpdA operons which express multilip-PDH complexes bearing one N-terminal lipoyl domain and up to ...

Wild type dihydrolipoyltransacetylase(E2p)-components from the pyruvate dehydrogenase complex of A. vinelandii or E. coli, and mutants of A. vinelandii E2p with stepwise deletions of the lipoyl domains or the alanine- and proline-rich region between the binding and the catalytic domain have been overexpressed in E. coli TG2. The high expression of A. vinelandii wild type E2p (20% of cellular pr...
